Experimental procedures and precautions for using Petri dishes

A petri dish is a laboratory utensil used for the culture of microorganisms or cells. It consists of a flat disc-shaped base and a lid, usually made of glass or plastic.

Petri dishes are delicate and fragile, so they should be cleaned and handled with care and care. The petri dish is best cleaned in time and stored in a safe and fixed location to prevent damage and breakage.

Experimental procedure

-Removal culture-medium

  1. Place the petri dish or culture plate on the side of the workbench.
  2. Start the air pump.
  3. Pick out the straw without cotton cold and poke it into the suction pipe of the air pump.
  4. Place the petri dish or culture plate in the center of the work area.
  5. Remove the lid from the petri dish or culture plate and place it on the back of the dish or culture plate, mouth up.
  6. Pick up the petri dish as close to the bottom as possible. Be careful not to touch the rim of the petri dish and do not pass your hand over the open.The computer generated the optional words: petri dish or its lid (with practice, the lid can be pulled off nicely without removing the lid completely,and the dish tilted to remove the old medium, which is faster and safer than the previous method).
  1. Tilt the petri dish and remove the medium. If no pump is available, discard the used medium into the waste beaker.
  2. Put the lid back on.
  3. Move the petri dishes to the opposite side of the work area from the untreated ones.
  4. Do the same with the remaining petri dishes and plates.
  5. Discard the straw and turn off the pump.

-Add culture medium or cells

  1. Place the necessary reagent bottles in place and loosen the lid of the reagent bottles to be used.
  2. Place the petri dish in the center of your work area.
  3. Remove the cap and suck the liquid from the reagent bottle with a straw.
  4. Put the lid of the petri dish behind the dish.
  5. Gently infuse one side of the petri dish directly into the base.
  6. Put the lid on the petri dish.
  7. Place the petri dish on the side where it was. Be careful not to let the medium get into the tiny space between the lid and the bottom.
  8. Repeat the steps to add medium to the second and remaining petri dishes.
  9. Ditch the straws.Again, with practice, you will be able to add the medium just as you would remove the cap

Petri dishes and porous culture plates are particularly vulnerable to contamination for the following reasons.

  1. The exposed surface area of the petri dish mouth is large.
  2. There is a risk of touching the edge of the petri dish when handling the open dish.
