Extraction and identification of plasmid DNA
Plasmids extracted by alkali lysis are separated according to the topological differences between covalently closed circular plasmid DNA and linear chromosome DNA. Within the narrow pH range of 12.0 to 12.5, the linear DNA double helix is unwound and denatured.
